LAB EQUIPMENT
- Stereoscopic microscope
- Fly Nap (Anesthetic)
- Fly Nap wands
- Isopropyl alcohol
- Jar
- Paint brush
- White index card
- Fly media (Carolina Biological Supply Company)
- 12 Yeast seeds
- 2 Culture viles
- 2 Vial plugs (sponge stoppers)
- 6 Wild type wings, wild type eyes - Female Fruit Flies
- 6 Apterous wings, sepia eyes - Male Fruit Flies
PROCEDURE
Day 1
Put four small yeast seeds in a vile with only the fly media in it. Take 4 to 6 flies out of the vile that was just female,these should all be (whatever phenotype they where) but should be check under the microscope. Then find 4 to 6 males out of the vile that contains both male and female( with what ever trait they had). The flies taken out of this second jar most be male or the experiment results will be wrong. The flies that you got out of those two viles into the vile that the yeast seeds where put into. Label this vile F1. Leave the flies in that vile to mate over the next few days.
Days 2-12
Look at the vile once a day until there are cocoons. Once there are cocoons the parents most be killed. To kill the parents use the fly nap and fly nap wand to put the fly parents to sleep. Once the flies have been knocked out put them into the “morgue”. The "morgue" is just jar with isopropyl alcohol in it and parchment paper on it as a lid. Keep coming to see what the vile looks like everyday and as the cocoons hatch take the flies out to see what the phenotype is. To see the phenotype use the fly nap the same why as when killing the parent, but before putting them into the "morgue" count the flies under the stereoscopic microscope and make note of the phenotype and gender then put them into the "morgue". Keep coming back everyday to do this until there are over 100 flies that have been counted. On the last day of counting F1 flies put 5 female and 5 male flies in a new vile and label it F2.
Days 13-25
Again look at the vile once a day until there are cocoons. Once there are cocoons the flies can be killed by being knocked out with fly nap and then put into the "morgue". Same as in F1 keep coming to see what the vile looks like everyday and as the cocoons hatch take the flies out to see what the phenotype is, take note of it, then put them into the "morgue". Do this until there are at least 100 flies that have been counted, the more flies the more accurate the experiment will be.
